Research Abstract

本研究は、ミトコンドリア翻訳系の異常に起因する疾患に着目し、その分子機構を解明することを目的とした。当研究ではこれまでに、MELAS、MERRF、MIDDなどミトコンドリア脳筋症由来のミトコンドリアtRNAに、コドン解読に重要なアンチコドン1字目の新規のtRNAタウリン修飾が欠損するという現象を見出している。MELASに関しては昨年度の研究成果により、主にwobble位修飾欠損により変異tRNAが、UUGコドンを翻訳できないということが判明した。この結果はMELAS発症の第一義的原因がtRNAの修飾欠損であることを意味している。また、UUGコドンの使用頻度が圧倒的に多い呼吸鎖酵素複合体IのサブユニットND6の欠損を示唆し、長年分子レベルでの理由が不明であった複合体I活性の低下というMELAS患者の生化学的特徴を説明できる。今年度は、患者検体由来の微量なRNAを用いて、高感度でタウリン修飾の有無を判別する方法を確立し、患者組織で実際にタウリン修飾欠損が生じていることを明らかにした。tRNALeu(UUR)遺伝子中に様々な点変異を有するミトコンドリア脳筋症の患者の骨格筋、心筋及び肝臓などの臓器から、微量な全RNAを抽出し、この方法を用いて解析を行ったところ、すでに培養細胞を用いた実験ですでに確かめている3243位と3271位の変異に加え、3244,3258,3291の変異を有する患者由来のtRNAにはタウリン修飾の欠損が明らかとなった。これら5つの点変異はすべてMELAS患者由来のものであった。一方、3242,3250,3254,3280位に点変異を有する患者由来のtRNAには修飾の欠損は観測されなかった。そしてこれら4つの点変異は、ミトコンドリアミオパチーやCPEOなど、すべてMELAS以外の臨床症状を示していた。これらの結果は、MELASがタウリン修飾欠損によって引き起こされる疾患であることを強く示唆すると共に、RNA修飾異常がヒトの疾患の原因になりうることを証明する結果であると考えている。
