Research Abstract

昨年度見つけ出した因子HMGA1aは孤発性アルツハイマー病(SAD)脳に特異的に観察されるプレセニリン2(PS2)スプライシング変種を産生させる原因となる因子で、PS2 pre-mRNA上の特徴的な配列に特異的に結合しており、低酸素刺激により核内での発現上昇(speckleへの蓄積)およびPS2 pre-mRNA結合能上昇、さらに、強制発現によって低酸素負荷を与えなくてもPS2Vを産生することを確認したので15年度はHMGA1aとPS2 pre-mRNAとの特異的な結合を制御することによってSADにおける細胞死を制御できるか否か検討した。また、HMGA1aが低酸素以外の酸化的刺激で誘導されPS2V産生に結びつく可能性を考え、アルミニウムを初めとする金属負荷によるPS2V産生についても検討を行った。その結果、HMGA1aが好んで結合するPS2 Pre-mRNA上の特徴的な配列をオリゴRNAによって作成し、低酸素前に細胞に導入しておくと低酸素負荷後のPS2V産生がオリゴRNAの導入量依存的に抑制できた。またその際、ツニカマイシン刺激のような小胞体ストレスによる脆弱性を回避することが可能だった。また、鉄、銅、アルミニウム、など金属による前処置により低酸素負荷時間を短縮してPS2Vの産生を観察することが出来た。さらに、細胞毒性を全く示さないようなアルミニウム濃度で長期間飼育していた細胞に低酸素負荷を施すと本来PS2V誘導に費やしていた10時間を僅か30分間で誘導出来ることが分かった。これらの結果はJournal of Neurochemistryはじめ各種雑誌に掲載予定である。
