Research Abstract

平成14年度に引き続き、動物肝臓や網膜にある水溶性酵素を生体内に存在するのに近い条件(生理的条件)の2次元電気泳動法で分離し、その酵素活性を網羅的に調べ、その知見を収集した。また、この2次元電気泳動法で分離された酵素の1次構造の解析をマトリックス支援レーザー脱離イオン化飛行時間型質量分析計やエレクトロスプレーイオン化タンデム型質量分析計を用いて網羅的に行い、その知見を蓄積した。これら収集した水溶性酵素の活性と1次構造の知見を対応させ、酵素活性と構造の情報を網羅した2次元電気泳動地図の作成を行なった。また、この地図を用いて、酵素活性阻害剤や薬剤などが酵素活性を阻害をする際に、それが特異的なのか、非特異的なのかを調べることができることを示した。これらの情報は国外の専門学術雑誌に公表した(Y.shimazaki, Y.Sugawara, T.Manabe.Proteomics (in press),2004:Y.Shimazaki, Y.Hiraga, M.Uesugi, T.Manabe.Biochimica et Biophysica Acta Vol.1696.p 51-57,2004:Y.Shimazaki, Y.Sugawara, Y.Ohtsuka, T.Manabe Proteomics Vol.3.p 2002-2007,2003)。
