Research Abstract

付着生物由来の接着物質の付着機構解明と工学利用を可能にするため、種々の海産二枚貝の接着タンパク質の化学合成および接着特性に関する研究を展開した。本年度では、海産無脊椎動物由来の材料の性質を分子レベルで解明する。海産生物由来繊維の分子構造を明らかにし、高分子化学的視点から用途開発を可能にするため、以下の研究を行った。
(1)足糸タンパク質のアミノ酸配列の決定
8種類の前駆体タンパク質のうち、最大高分子量のタンパク質の精製およびアミノ酸配列分析を行った。このタンパク質は、10残基のペプチドが繰り返して配列する構造タンパク質の一つであることを明らかにした。このタンパク質はポリフェノール性タンパク質の一つであると考えられる。10残基のうち2残基は、構造不明なアミノ酸であったが、他の分析結果から、少なくとも一つには糖鎖が結合していることが明らかになった。
(2)イガイ足糸繊維タンパク質の分析
足糸を形成する繊維素タンパク質を全て同定し、各々の構成タンパク質がどのように配置され、繊維の強度向上にどのように寄与しているかという問題に取り組むため、はじめに、足糸を形成するタンパク質の生化学的分析を行った。その結果、足糸は、複数種類の前駆体タンパク質がキノンの縮合反応に基づく架橋反応によって形成され、少なくとも8種類、内、一つは糖鎖が結合しているタンパク質であることが明らかになった。
(3)イガイ類足糸を模倣した高強度繊維材料の開発
海洋接着タンパク質はコラーゲンと複合して高次階層構造を形成し、極めて強い物理的強度と安定な化学的・生化学的特性を持つ。接着タンパク質と天然多糖の複合材料系を研究し、バイオミメティックな繊維材料の作成を試みた。イガイ足糸を模倣した高強度繊維の開発のため、イガイの足糸形成機構に基づき、ポリペプチドの分子を設計し、実際に、合成、繊維化、および機械的特性の評価を行った。
