Research Abstract

1)新規GA非感受変異体の選抜-名古屋大学に系統保存されている矮性変異体ライン、九州大学においてMNU処理で見いだされた矮性変異体ライン、Tos17で誘発された矮性変異体からGA変異体に特徴的な表現型(矮性、濃緑葉、幅広な葉)を指標にGA関連矮性変異体の選抜を試みた。表現型による1次選抜後、選抜個体にGAを投与しGAの反応性を調査し10個体のGA非感受変異体候補を選抜した。
2)SLR1ホモログ(SLRL1,SLRL2)の分子遺伝学的解析-これまでにGA情報伝達の抑制因子SLR1を同定し、分子遺伝学・生化学的な解析を行ってきた。その研究過程でイネゲノム中に比較的相同性の高い2つのSLR1ホモログ(SLRL1,SLRL2)を見いだした。SLRL1&2とGA情報伝達の関与を調査したところ少なくともSLRL1とGAの部分的な関与が示唆された。
3)マイクロRNAを介したGAの情報伝達の分子遺伝学的解析-GAmybはGA情報伝達に関与する転写因子である。我々はイネゲノム中にはGAmybの配列の一部に完全マッチするマイクロRNA(miR159)を見いだした。このマイクロRNAとGA情報伝達との関係を明らかにするために分子遺伝学的解析を行った。miR159を過剰発現した個体ではgamyb変異体と同様に節間伸長や花器の形成に異常を来したことからこのマイクロRNA(miR159)がGA情報伝達に寄与していると結論した。
4)GAmybを介したGA反応遺伝子の解析-gamyb変異体の胚乳を用いてマイクロアレイ解析を行った。その結果胚乳においてGA依存的な遺伝子の転写はすべてGAmybを介して制御されていることが明らかになった。
5)GA情報伝達因子GID1とGID2因子の分子遺伝学・生化学的解析-GA情報伝達因子であるGID1およびGID2の生化学的な特性について明らかにした。
