Research Abstract

昨年度までに、アガサレジノールのような低分子化合物の免疫電子顕微鏡観察を目指すとき、その化合物が、試料調製(植物組織の固定・脱水・包埋、超薄切片の調製、および超薄切片の抗血清処理)において本来の存在場所から移動し、さらに組織外へ溶脱した可能性が示され、この問題の対策として、(1)低温下・短時間での試料調製、(2)試料の凍結乾燥、(3)試料のカフェイン溶液処理、を試みた。しかしながら、免疫標識の十分な改善は達成されなかった。本年度、「前固定免疫標識法(あらかじめ低分子量物質と抗体タンパクとを結合させる、タンパク質の固定は比較的容易である)」を試みた。この方法では、Fab断片の調製が必要となるため、その調製を行った。まず親和クロマトグラフ法によって抗アガサレジノール抗血清からIgG画分を分取し、続いてこの画分をパパインで酵素分解し、分解物から親和クロマトグラフ法によってFab断片を単離した。得られたIgG画分およびFab断片の抗体活性を免疫検定した。その結果、IgG画分は認識選択性は向上したものの、活性自体は低下し、Fab断片に関しては濃度が極めて薄く、活性も低下した。これらのIgG画分およびFab断片は、免疫組織化学的検討には不適当であり、血清の精製方法を検討する必要がある。
