Research Abstract

産業動物であるニワトリにおいて体重の増加・成熟期間の短縮化という課題を改善することは、飼育に伴う維持管理・飼料コストの節約、食肉・食卵の増産を可能にする。この課題は経済動物の究極的目標であといえる。成長ホルモン-インスリン様成長因子は体重の増加や産卵効率に関わる因子である。これまでに生長ホルモン(GH)受容体(GHR)-インスリン様成長因子(IGF)という作用経路を明らかにし、この機構を明らかにするため、IGFの作用調節因子であるIGF結合蛋白(IGFBP)-1,4(ACCESSION No.AY230206) cDNAの全塩基配列を決定し、ゲノム遺伝子の構造、塩基配列配列を明らかにした。さらにIGF・IGFBP-4の主な合成器官である肝臓でのIGF、IGFBP-4mRNA合成はGHに依存的に合成されることをin vivoで明らかにした。現在これを用いて転写発現機構を解明している。またこれらの抗体を作成し、その力価を検定した。in vivoにおいて個体、組織の成長における様々な生理状態;胚発生期、成長段階、性成熟期、GHR欠損矮性鶏、甲状腺ホルモンの阻害等の実験を行い、IGFBP-4が成長ホルモンによって調節されていることを明らかにした。今後、上述の情報をもとに遺伝子の転写発現機構をさらに明らかにする。
