| Research Abstract |
1.PCM-1のドミナントネガティブ変異体によるcentriolar satellitesの機能・構造解析
PCM-1蛋白はcentriolar satellitesのscaffold proteinであることをこれまでの研究で示してきた。今回PCM-1蛋白同士の結合を担う部位を同定した。このPCM-1同士の結合を担う部分のペプチドを細胞に高発現させるとcentriolar satellitesを分解する事ができた。このペプチドによりendogenousなPCM-1同士の結合が阻害される事でcentriolar satellitesが分解すると考えられる。Centriolar satellitesが分解された細胞はPCM-1をRNAiによりknock-downした細胞とほぼ同じphenotype(中心体のpericentrin量の減少、細胞周期停止)を示した。これらの結果から、PCM-1蛋白がcentriolar satellitesという構造を取ることが、その機能発現に必須であることが示された。
2.ツーハイブリ法によるPCM-1結合蛋白の探索
PCM-1蛋白の各ドメインをbaitとしてツーハイブリ法を行い、PCM-1結合蛋白を探索した。いくつかのcandidateについて、タグをつけて細胞に発現させ、その局在を検討した。また、可能性のあるものについて抗体を作成し、細胞内局在およびPCM-1との結合を検討中である。
