骨外性粘液型軟骨肉腫における微小管及びその関連蛋白の解析

Research Abstract

本研究では,まれなヒト軟部肉腫の一つであり,未だその組織起源について明らかにされていない骨外性粘液型軟骨肉腫がしばしばその腫瘍細胞の細胞質内に特徴的な微小管の集合像が認められることに着目し,本腫瘍の神経ないし神経内分泌細胞への分化の可能性について調べることを目的として検索を行った。本年度は,収集しえた25例の骨外性粘液型軟骨肉腫のホルマリン固定パラフィン包埋腫瘍組織を用いて,神経細胞ないし神経細胞から派生する細胞に特異的に分布するとされているmicorotubules-associated protein 2(MAP-2)とclass III β-tubulinの発現を免疫組織化学的に調べた。その結果,MAP-2は21例(84%),class III β-tubulinは13例(52%)に発現がみられ,その発現の程度は各腫瘍によりさまざまであった。次に,同検体についてClass III β-tubulinに特異的プローブを用いたin situ hybridizationを行い,2例にclass III β-tubulin転写産物の発現が認められた。更に,通常の電顕的検索において腫瘍細胞の細胞質内に微小管の集合像が観察された1例でclass III β-tubulinの金コロイド標識による免疫電顕を行った結果,微小管に一致してコロイド粒子が観察された。以上のような結果より,骨外性粘液型軟骨肉腫に観察される微小管は神経細胞のそれと類似するものであり,本腫瘍の一部は神経ないし神経内分泌細胞への分化する可能性があることが示された。
