Research Abstract

[背景]チェルノブイリ原発事故汚染地域(チェ地域)の甲状腺乳頭癌(PTC)でのβ-catenin胞体内発現やcyclin D1過剰発現への、β-catenin遺伝子(CTNNB1)とAPC発現の関与を検討した。Pin1がcyclin D1転写を調節することが示され、その発現も検討した。[方法](1)チェ地域のPTC凍結組織14例より核酸を抽出しCTNNB1 exon3の変異をPCR-SSCPとシークエンスで解析(2)チェ地域の濾胞腺腫(FA)とPTC各14例のcyclin D1転写レベルをreal-time PCRで解析、β-catenin発現との相関を検定(3)PTCのパラフィン切片上でのAPC発現を免疫組織化学的に検討(4)FAとPTCでのPin1発現をRT-PCRと免疫組織化学により解析し、β-catenin発現との相関を検定[結果](1)PTC14例にCTNNB1 exon3の変異はなかった。(2)cyclin D1転写過剰発現はFAの45.5%、PTCの54.5%であった。β-catenin胞体内発現はFAの85.7%、PTCの100%に認めたが、FAでは全例弱陽性であった。cyclin D1と胞体内β-cateninの発現レベルに有意な相関を認めた(p=0.045)。(3)PTC全例にAPC蛋白の発現を認めた。(4)Pin1の過剰発現はFAの28.5%、PTCの100%で、PTCのPin1発現はRT-PCRで確認した。Pin1と胞体内β-cateninの発現レベルに正の相関を認めた(P<0.001)。[考察]FAにcyclin D1 mRNA過剰発現を認め、甲状腺腫瘍のcyclin D1転写活性の亢進は早期のイベントであるのかも知れない。PTCのβ-catenin胞体内蓄積とcyclin D1過剰発現への、CTNNB1 exon3変異やAPC蛋白発現低下ではない、Pin1を介したWnt伝達系活性化の関与を示唆する。
