Research Abstract

正常線維芽細胞ではSmad3を強発現した場合、無刺激下およびTGF-β刺激下のコラーゲン遺伝子転写活性は有意に亢進したがSmad2、あるいはSmad4を強発現にて特に変化は認められず、強皮症線維芽細胞ではこれらSmad蛋白の強発現にて無刺激下およびTGF-β刺激下のコラーゲン遺伝子転写活性は特に変化は認められなかった。Sp1の強発現は、正常維芽細胞ではTGF-β刺激に対する反応性を亢進させたが、強皮症線維芽細胞では変化しなかった。Smad3とSp1の強発現は、正常皮膚線維芽細胞では無刺激下およびTGF-β刺激下のコラーゲン遺伝子転写活性は著明にに亢進したが、強皮症皮膚線維芽細胞では軽度亢進し、これらの結果は強皮症皮膚線維芽細胞ではautocrine TGF-β signalingにて既に活性化しているためと考えられた。またこの考えに合致して、強皮症皮膚線維芽細胞ではSmad3のセリンリン酸化が亢進し、Smad3とp300およびSp1の結合が亢進していた。強皮症線維芽細胞ではSmad7発現が亢進しTGF-β受容体と恒常的に結合していることが示された。Smad7の発現は正常線維芽細胞ではコラーゲン遺伝子転写活性を抑制したが、強皮症線維芽細胞では変化がなく、Smad7によるnegatvie feed back機構に異常があると考えられた。強皮症線維芽細胞では恒常的にp38 MAPKが活性化し、P38 MAPキナーゼ特異的阻害剤(SB202190、SB203580)あるいはdominant negatvie p38 MAP kinase強発現にて強皮症繊維芽細胞における細胞外マトリックス産生亢進が抑制され、強皮症線維芽細胞における細胞外マトリックス過剰産生においてp38 MAPKが関与する可能性が示唆された。
