Research Abstract

アトピー性皮膚炎(AD)はTh2優位の皮膚アレルギー性疾患である。今年度は、Th2タイプのケモカインであるTARC/CCL17と、その受容体であるCCR4と、Th2タイプのサイトカインであるGM-CSFの遺伝子多型解析をAD患者を対象に行った。
TARC遺伝子に関しては、promoter領域(-431C/T)に遺伝子多型を昨年度見い出しているが(Sekiya et al. Immunogenetics 54:742-5,2003)、今年度はこの多型がTARC遺伝子のpromoter活性に影響していることを証明した。しかし、AD患者においてこの遺伝子多型との明らかな相関は認められなかった(Tsunemi et al. Exp Dermatol 13:715-9,2004)。
CCR4遺伝子多型に関しては、1014C/T多型が知られている。AD患者を対象に1014C/T多型を解析したが、有意な相関は認められなかった。喘息合併の有無、ADの重症度、血清IgE値、末梢血好酸球数などにも注目して解析したが、やはり有意な相関は得られなかった(Tsunemi et al. Acta Derm-Venereol 84:187-90,2004)。
GM-CSF遺伝子に関しては、intron3(3606T/C),exon4(3928C/T)に遺伝子多型が知られており、カナダのAD小児患者において、これらの多型との有意な相関が報告されている。そこで本研究でもAD患者を対象に3606T/C,3928C/T多型を解析したが、有意な相関は認められなかった。
