Research Abstract

CRF R2βを自然発現しているA7r5血管平滑筋培養細胞に、CRF, Ucn, Ucn IIなどのCRF family peptidesを添加し、IL-6 ELISAシステムを使用して、IL-6濃度を測定した。IL-6濃度の増加は、Ucnで最も増加させ)、Ucn IIでも増加作用を認めたが、CRFではほとんど認めなかった。このことから、血管平滑筋培養細胞ではUcn又はUcn IIがCRF R2βを刺激して、cAMP経路を活性化することでIL-6分泌作用をもたらすと考えられた。
Ucnは、心血管系に発現するCRF R2βを介して、心陽性作用や血管拡張作用を示す。ラット血管のモデルを使用し、UcnやUcn IIの血管拡張作用と伝達経路を調べたところ、同ペプチドでの血管拡張作用を認め、protein kinase A (PKA)経路を介しているとわかった。MAP kinase (MAPK)経路の関与も示唆された。
更に、CRF R2β mRNAの発現レベルの調節に関わる細胞内伝達機構について解明した。Ucn等のCRF R2βのagonistによる同受容体の刺激は、cAMP経路の活性化の作用によりCRF R2β mRNAの発現レベルを低下させた。また、そのcAMPによる発現調節の一部はPKA経路を介しているとともに、MAPKの関与も示唆された。
下垂体ACTH産生腫瘍において、内分泌学的に自律過剰分泌を証明できるが、臨床的には特徴を示さないクッシング病をプレクリニカルとして報告した。この報告はクッシング病の病態を推測する上で役に立ち、早期発見される上で役立った。
更に、CRF R2β1/2のサブタイプや下垂体でのCRF R2の発現を発見して報告した。
