Research Abstract

本研究においてはヒト口腔扁平上皮癌細胞株B88細胞を用いて抗癌剤および放射線照射によるアポトーシスの誘導と転写因子NF-κBの関連について解析し以下の結果を得た。
NF-κBの腫瘍細胞に及ばす影響を検討するため、B88細胞に変異型IkB-α cDNAを遺伝子導入し、安定的にNF-κB活性を抑制した細胞クローン(B88mI)を作成したB88細胞とB88mI細胞のin vitroでの細胞増殖能を比較したところ、両細胞間に明らかな増殖能の差は認められなかった。一方、これら細胞クローンをヌードマウス背部皮下に移植し、各々の造腫瘍能につき比較検討すると、NF-κB活性を抑制したB88mI腫瘍の造腫瘍能はB88腫瘍と比較して有意に低下していた。次に抗癌剤5FUにて両細胞クローンを処理するとB88mI細胞はB88細胞と比較して7-23%の細胞増殖抑制効果を示した。また、放射線照射によりB88mI細胞はB88細胞と比較して16-46%の細胞増殖抑制効果を示した。尚、フローサイトメトリーの検索より、この細胞増殖抑制がアポトーシスの誘導により生じたことを確認した。更に坦癌ヌードマウスに5FU(15mg/kg)を腹腔内投与あるいは放射線照射(total 12Gy)を行った結果、B88腫瘍では有意な腫瘍縮小は認められなかったが、B88mI腫瘍においては、抗癌剤および放射線照射ともに明らかな腫瘍の消失を認めた。
我々の以前の研究結果よりヒト唾液腺癌細胞を5FUで処理するとNF-κB活性の抑制が認められ、更に抗アポトーシス蛋白(TRAF-2,cIAP-1)の発現低下とアポトーシス関連因子(Caspase8,Caspase3)の活性化が認められたことから、本研究においてB88mI細胞が抗癌剤と放射線照射により高い腫瘍抑制効果を示したのは、NF-κB活性の抑制がアポトーシス誘導を促進したものと考えられる。
