Research Abstract

平成14年度に引き続き,癌転移に対し促進的に作用する過剰な活性酸素種や活性窒素種などの酸化ストレスの消去系亢進の指標として,NO産生抑制活性を一次スクリーニングとして実施したところ,新たに漢薬"小紅参"(Rubia yunnanensis,根部)およびエジプト産天然薬物Anastatica hierochuntica(全草)の抽出エキスに活性が認められたことから,活性を指標に分離,精製し,有効成分を探索した.その結果,小紅参から新規化合物としてarborinane型トリテルペンrubianol-a-g, arborinane型トリテルペン配糖体rubianoside I-IVおよびアントラキノンrubianthraquihoneを単離,構造決定するとともに,含有環状ペプチド配糖体RA-XII (IC_<50>=0.85μM)およびそのアグリコン部であるdeoxybouvadin (IC_<50>=0.015μM)に顕著なNO産生抑制活性を見出し,その活性強度はチロシンキナーゼ阻害剤であるherbimycin A (IC_<50>=0.094μM)よりも強い活性であった.また,RA-XIIおよびdcoxybouvadinの作用点のひとつにiNOSの誘導を抑制することを明らかにし,これら環状ペプチド類が本課題の候補化合物として有望であることが示唆するなどの結果を得た.次いで,我々が保有するflavonoid化合物ライブラリー73種のNO産生抑制活性スクリーニングを実施し,活性発現の必須構造や構造活性相関を明らかにするとともに,漢薬"木香"の含有セスキテルペン成分やそのアミノ酸付加体に,NF-κBの活性化や熱ショック蛋白(HSP-72)の発現抑制作用を認めるなど,NO産生抑制作用メカニズムに関する知見を得た.
上記の手法を用いピックアップした候補素材のうち,日本薬局方生薬の"川骨"(Nuphar pumilum,根茎)MeOH抽出エキスおよび含有成分について,B16メラノーマ4A5細胞を用いたコラーゲンコートメンブランにおける癌細胞浸潤抑制活性(in vitro)およびマウスを用いた癌細胞転移抑制作用(in vivo)試験を実施した.その結果,エキスのアルカロイド分画および含有成分である含硫黄セスキテルペンアルカロイド2量体6-hydroxythiobinupharidineに顕著な癌転移抑制作用を見出し新たな医薬シーズの探索に成功するなど,平成15年度の当初計画をほぼ達成した.
