Research Abstract

Pin1は細胞周期調節因子として発見された。すなわちCdc2/サイクリンBの脱リン酸化を行うcdc25cのリン酸化部位に結合しその立体構造を変化させることで活性を調節している。その後Pin1と結合する分子としてP53,c-Myc, Jun/Fosなどの転写因子調節タンパク質が同定され、Pin1はそれらの活性調節を行っていることが判明してきた。私はPin1ノックアウトマウスを作成し解析を行ったところ、Pin1分子単体の欠損によっても生育可能であったことから代替の分子が存在すると仮定し、その分子の探索を行った。その結果Par14分子の発見につながった。Par14はPin1同様にペプチド中のプロリンをシス体からトランス体に異性化するPPIase活性を有しており、Pin1機能の類推に役立っている。また、ノックアウトマウスより得た細胞を用いた実験により、Pin1が結合する分子として新たにいくつかの分子の同定に成功した。たとえば、CREBは転写の活性化に133番目のセリンのリン酸化が必要であるが、CREBタンパク質中のセリン(リン酸化)-プロリンと並ぶ領域のいくつかにPin1が結合し、CREBの転写活性を調節していることを発見した。以上のようにPin1の分子メカニズムとしての転写活性調節機能を明らかにしてきた。また、本研究を行うにあたり、機能解析のためにいくつかの実験系の開発も行ってきた。例えば、従来型の転写因子の活性測定に用いられていたゲルシフトアッセイやルシフェラーゼアッセイなどの方法は、単一の転写因子の解析はできても複数の転写因子の解析を同時に行うことは困難であったが、マイクロアレイ技術を用いた蛍光プローブを転写因子解析に用いることで、一度に50種類以上の転写因子の活性化状態を検出できる系を開発した。すなわち、Pin1の機能解析と新規転写因子解析手法の両方を満足させた。
