Research Abstract

昨年度までの段階でマウスES細胞由来奇形腫中に確認された、中腎管様あるいは尿管芽様構造の解析を進めた。今年度新たに複数のマウスES細胞株において再現性を評価した。
従来の市販ES細胞株(129sv系マウス由来)に加え、ES-D3株とR1株を新たに導入し、腎臓発生必須遺伝子発現および奇形腫内腎臓原基構造の検出について追加実験を行った。RT-PCR解析による胚様体発生過程でのPax-2、lim1、WT1、Sall1、Eya1、GDNF、c-Ret、Emx2、Wnt-4の発現パターンは、3つのES細胞株間で近似しており、再現性が確認された。特に腎臓発生の最初期に発現するPax-2が、胚様体接着後6-9日に発現することから、腎臓原基細胞の出現は早くともこの時期以降と推察された。
移植14日、28日の時点で組織化学的手法による腎臓原基構造の探索を行った。両時点において胚様体由来奇形腫内に、Pax-2陽性・Ksp-cadherin陽性の分枝管状構造を検出した。どのES細胞株においても、これらの管状構造は、中腎管あるいは尿管芽に極めて類似した組織化学的・形態学的特徴を有し、また中腎ネフロン類似の曲細管も認められた。移植28日では、一部にPax-2陰性のKsp-cadherin陽性管状構造が検出され、生体内でのこれら構造の分化が示唆された。
一連の研究により後腎発生に繋がる中腎管・尿管芽系細胞が、ES細胞から分化し得ることが明らかとなり、腎臓再生研究の基盤となるものと考えられる。研究成果の一部は、現在Am J Physiol-Renal Physiolに投稿中である(2^<nd> revision)。また、今回、後腎ネフロンの形成は組織化学的に確認できなかったことから、造後腎中胚葉については、積極的な分化誘導が必要と考えられ、今後の大きな課題として残る。
