Outline of Annual Research Achievements

生体酵素である、Prostaglandin endoperoxide H synthases (PGHSs)のバイオミメティック触媒である、ポルフィリン錯体("ツインコロネット"型錯体)によって触媒されるジエンの位置選択的および立体選択的酸素化のメカニズムを理論化学計算で調べることを目標とし、さらにPGHSsの反応機構を理解することを試みた。手法としては、two-layer ONIOM (QM/MM)法及びAFIR法を用いた。特に、溶液中で最も熱力学的に安定な反応前駆体、中間活性種の生成過程、位置選択的水素の引き抜き反応、そして、ジエンの酸化過程について、詳細に調べた。選択的過程については、遷移状態構造の系統的かつ効率的な探索をAFIR法を応用し実行した。また、この錯体は300原子を超える系であり、応用する分子力場の依存性が高いことが考えられる。そのため、我々は、様々な種類の分子力場(AMOEBA09[分極可能],MMM3, MMFF, OPLS-AA, CHARMM22)を適用し、反応経路などに違いを詳細に検討した。その結果、反応経路の概略および位置選択性の決定過程が解明された。
また、ノンヘム鉄(III)-スーパーオキソ系においては、中間体を含めた詳細な電子構造の解析とともに、2-ヒドロキシ2-アザマンタン、フェニルヒドラジンを基質とした、水素引き抜きの反応性について調べた。方法は、上述のtwo-layer ONIOM(QM/MM)法を使用した。その結果、反応機構の概略を解明した。
