Outline of Annual Research Achievements

H27年度の実績概要として以下の3点があげられる。
1点目は、好アルカリ菌(以下、TA2.A1)由来のATP合成酵素のF1部(以下、TA2. F1)の一分子回転計測である。F1部はATP合成反応における触媒部位を持つため、その基本特性の詳細な解析(速度論的パラメーターや回転トルク等)は重要な意味を持つ。まず、一分子回転計測で使用する変異体の最適化を行った。次に、顕微鏡下で200 nmのビーズを用いた回転計測からTA2. F1のATP加水分解反応における詳細な基本特性の解析を行った。最大回転速度は約6.5Hz、Kmは2.7 uM、ATP結合速度は9.9x10^6/Msであった。また、TA2. F1は他生物由来と比較して大きい回転トルク(53 pN nm)を発生する事から、他生物由来より強固な化学力学共役機構を持つことが示唆された。これらの結果は原著論文として出版された。
2点目は、ATP合成酵素にH+駆動力を提供するcytochrme bo3複合体の生化学機能評価である。cytochrme bo3を再構成した人工膜小胞を用いて、cytochrme bo3の持つユビキノンの酸化活性に共役したH+輸送活性を評価できる電気計測系を開発した。これによりcytochrome bo3の持つH+輸送の詳細な解析を行った。現在、一分子cytochrme bo3のH+輸送計測を試みており、より詳細な輸送機構の解明を行う。
3点目は、病原菌(以下、Fuso)由来のF1の一分子回転計測である。発現系・回転計測系を構築し一分子回転計測を行ったところ、他生物種由来よりも極めて速い~1000Hzの速度で回転することがわかった。さらに、FusoF1は極度に阻害状態に陥ることがわかった。今後更なる解析を行なっていく。
