Outline of Annual Research Achievements

マツ材線虫病の主病原体はマツノザイセンチュウBursaphelenchus xylophilusであるが、そこに取り巻く細菌の種類やその特徴について、すなわちマツノザイセンチュウ体表面、ベクターであるカミキリ日本産Monochamus alternatusおよびポルトガル産M. galloprovincialisの消化管と気管内、健全木および本病による枯死木樹から名井に存在する細菌の種構成について、分離培養法と16SrDNA のV4 領域をもとにしたメタゲノム解析法の2つの方法により調べることができた。その結果、(1)マツ樹木を主たる生活の拠り所とする線虫とカミキリとを併せて、3者間でマイクロバイオームが共通していること、(2)本病により枯死したマツ樹体内のマイクロバイオームがより多様性を増すことを示せた。そのうちの培養法により選抜した細菌2種を選び、完全ゲノム配列を解析したところ、(3)植物内生菌(endophytic bacteria)グループに属し、解毒代謝機構が非常に発達していることが分かった。最後に、クロマツ苗を用いた人工感染試験を実施したところ、(4)病原性を有するマツノザイセンチュウが樹体内に侵入する際に、単独では病原性の無かった内生菌が、病原性を助長する能力を持ってしまうことを示すことができた。
マツノザイセンチュウは北米原産のため、東アジアおよびヨーロッパにおいては外来種である。病原性を持ったマツノザイセンチュウが樹体内に侵入すると、共生関係を確立していなかった在来マツは病原体と様々な攻防を経て枯死に至ることが報告されてきたが、今回、マイクロバイオームの変遷とその機能解析というこれまでにない新しい視点から、本病のメカニズムを説明することができた。
