Outline of Annual Research Achievements

本研究課題は,テラヘルツ分光を用いてキセノンが圧入された際に生じる水分子の構造化を評価し,植物中における水の構造化が細胞の生理状態に与える影響に新たな知見を見出すことを目的としている.平成28年度は,細胞の分光測定に適した全反射減衰テラヘルツ時間領域分光(THz TD-ATR)測定系を自作し,それを用いてヒト由来のHeLa細胞中の細胞内水を詳細に評価できることを示した.しかし,植物細胞にキセノンを圧入し,細胞内水の構造化を評価するには至らなかった.
テラヘルツ帯の複素誘電率はブロードな誘電応答の重ね合わせで表現されるため,水の動態・構造に関する詳細な情報を正確に得るためには広帯域にわたる分光測定が必要不可欠である.そこで本研究では超短フェムト秒パルスを用いたTHz TD-ATR測定系の自作を行い,0.2~7.0 THzまで測定領域を広帯域化することに成功した.このTHz TD-ATR測定系の妥当性検証のため270-340 Kの温度範囲で純水の複素誘電率測定を行ったところ,昇温に伴って水の複素誘電率は大きくなる傾向が認められ,さらに270 Kの過冷却水が相転移して氷Ihに変化すると複素誘電率が非常に大きく変化するという結果が得られた.これらの結果は先行研究の報告例ともよく一致しており,本測定系で得られる複素誘電率が妥当であることを確認することができた.基板に接着して分裂し,細胞単層を形成するHeLa細胞はTHz TD-ATR測定に適した細胞試料であるため,構築したTHz TD-ATR測定系を用いてHeLa細胞の複素誘電率測定を行い,その結果から細胞内の水和状態ならびに水素結合環境の定量評価を行った.その結果,HeLa細胞内の水の約25 %が水和水として存在しており,残りの75 %の細胞内水は通常の水に比べて極めて無秩序な水素結合構造を形成していることが示された.
