希少脂肪酸生産に有用な微生物の探索と機能解析

Outline of Annual Research Achievements

EPAやDHAを変換する微生物の探索により、EPAやDHAを変換する様々な微生物の取得に成功した。EPAを変換する菌株のEPA変換物の構造を解析したところ、本菌の生成物は、共役二重結合を有するcis-5,cis-8,cis-11,trans-13,cis-17-eicosapentaenoic acid、cis-5,cis-8,trans-11,trans-13, cis-17-eicosapentaenoic acidおよび、部分飽和化されたcis-5,cis-8,trans-13, cis-17-eicosatetraenoic acidであることを明らかにした。さらに、本菌株の16S rDNA解析により、本菌がClostridium bifermentansであることを明らかにした。また、本菌の基質特異性の解析から、本菌は炭素数18および20の不飽和脂肪酸のω6位とω9位のシス型二重結合を認識し、ω7位とω9位に二重結合を有する共役不飽和脂肪酸に変換し、さらにω9位の二重結合を飽和化することを明らかにした。 C. bifermentansの洗浄菌体を用いて反応条件の検討を行ったところ、0.8 mMのEPAから0.5 mMの共役EPAを生産することに成功した。また、部分飽和化EPA生産に適した反応条件を検討したところ、0.8 mMのEPAから0.5 mMの部分飽和化EPAを生産することに成功した。また、Micrococcus luteus由来の水酸化脂肪酸脱水素酵素を発現する形質転換大腸菌を作成し、さらに、NAD+再生系とカップリングすることで、各種10-ヒドロキシ脂肪酸や12-ヒドロキシ脂肪酸(約1 M、300 g/L)から対応するオキソ脂肪酸を変換率90%以上で生産することに成功した。これらのEPA変換物やオキソ脂肪酸は希少脂肪酸であり、これまでに供給源が存在しなかった希少脂肪酸を生産することに成功した。
