本研究では、葉緑体のイソプレノイド代謝をモデルケースとし、細胞小器官内部での基質の局在化のメカニズムを、酵素の局在に着目し、明らかにすることである。申請者は、様々な事実から、LIL3というタンパク質が、geranylgeranyl diphosphate (GGPP)からphytyl diphosphate (PPP)へと還元するgeranylgeranyl reductase (GGR)をチラコイド膜につなぎ止めることによって、GGPPとPPPの局在を葉緑体内で制御しているのではないかとの仮説を立てた。この仮説を検証するため、第一にGGPPおよびPPPの測定方法の確立を目指した。また、GGPP、PPPの基質の局在の違いが、LIL3によるものであることを検証するため、LIL3の局在をチラコイド膜から包膜に変えた形質転換植物を作成した。また、申請書には記載していないが、LIL3がGGRとどのように相互作用しているのか、さらなる知見を得るために、シングルパーティクル（単粒子）解析を試みた。
