Outline of Annual Research Achievements

これまでの結果より、定常状態における血管内皮細胞を経由した抗体移行は血管壁のバリア機能によって大きく制限され、定常状態では、抗体は血管内皮細胞間隙からはほとんど移行せず、血管内皮細胞内の細胞質を介するトランスサイトーシスの経路によって皮膚へ移行すると推測された。過去の報告より、fluid-phase pinocytosisによる血管内皮細胞のIgG取り込みが行われると記載され、トランスサイトーシスのinitiationと考えられているが、その詳細なメカニズムは解明されていない。有核細胞による細胞外物質の取り込みは、マクロピノサイトーシス、クラスリンによるエンドサイトーシス、カベオラによるエンドサイトーシスなどに大別される。このうち、血管内皮にはカベオラ小胞、クラスリン小胞が豊富であるという過去の知見から、IgGの取り込みはクラスリン小胞、カベオラ小胞のいずれかの経路によると予測した。まず、蛍光免疫染色により、血管内皮細胞に取り込まれたIgGと、カベオラ、クラスリンの構成タンパクであるカベオリン-1とクラスリンと共局在するかを検討したところ、カベオリン-1とより多くのIgGが共局在して存在することが明らかとなった。また、in vitroにおいて、血管内皮細胞のIgGの取り込みは、カベオラを構成する脂質ラフトの形成を阻害するNystatin、あるいはカベオリン-1に対するsiRNAによって減少することが明らかとなった。さらに、c-Ablがカベオラ機能に重要であるという報告を踏まえ、c-Abl阻害薬であるimatinibをマウスに投与し、実験系1を用いて抗体の皮膚移行に影響するかを検証したところ、imatinibの投与量に応じて抗体の皮膚移行は減少することが明らかとなった。
