Outline of Annual Research Achievements

免疫記憶は獲得免疫機構の根幹をなす機能である。近年ヒトおよび霊長類において、新規メモリーT細胞集団であるステムセルメモリーT細胞 (TSCM) が発見された。TSCMはナイーブT細胞マーカーを有するメモリーT細胞集団である。またTSCMは幹細胞性や長期生存能を有し、免疫記憶の根幹を担う重要なメモリーT細胞集団であると考えられている。しかしながらTSCMがどのようなメカニズムで誘導されるかはあまり明らかになっていなかった。
一方で近年のがん研究において細胞移入療法は従来の抗がん剤をしのぐ、非常に有効な治療法として考えられてきている。TSCMをがん治療に応用することを目的として、我々はまずin vitro によりTSCM様細胞(iTSCM)を誘導する方法を確立した。CD4+およびCD8+活性化T細胞を抗原非存在下でNotchリガンドを発現するフィーダー細胞と共培養を行った。共培養の結果、一部の活性化T細胞はナイーブT細胞マーカーを示す細胞(iTSCM)へと分化した。CD4+およびCD8+ iTSCM を担癌マウスに移入したところ、ナイーブT細胞やエフェクターT細胞、TCMに比べて有意に腫瘍の増大を抑制した。次にヒト活性化T細胞を、Notchリガンドを発現するフィーダー細胞と共培養を行った結果、マウスと同様にiTSCMが誘導された。さらにヒトiTSCMのミトコンドリアでは酸化的リン酸化が亢進し、非常にenergeticな細胞であることが明らかになった。
以上の結果から私は学術振興会特別研究員採用第3年度目において、NotchシグナルがTSCMを誘導し、誘導されたTSCMがマウスおよびヒトにおいて強い抗腫瘍活性を有することを示した。これらの研究結果はNature Communications誌に掲載予定である。
