Outline of Annual Research Achievements

過去２年間の研究から、Sim1という遺伝子が風切羽の形成領域において鳥類に特異的な発現を示すことが明らかとなっていたため、当該年度は、Sim1と風切羽の関係性の詳細な調査を中心に研究を展開した。
本来ニワトリの後肢はうろこで覆われているが、この領域に羽毛をもつ品種が存在する。そこで、この品種を用いて風切羽とSim1の発現の有無を調査した。まず成鳥の後肢を観察すると、その一部に風切羽が生えていることが判明し、さらに胚発生期においてその領域にSim1の発現が見られた。これらの結果は、Sim1の発現が風切羽形成に関与することを示唆するものである。なお、これらの解析は、ナショナルバイオリソースプロジェクト（ニワトリ・ウズラ）の支援を受けて遂行された。
また、Sim1が風切羽へと神経軸索を誘因する機能をもつ可能性に着目し、これを検証した。野生系統のニワトリ胚の前肢および上記の脚羽品種胚の後肢における神経配向の状態を可視化したところ、Sim1の発現箇所に向かって軸索が伸長している様子が観察された。一方、Sim1が発現しない野生系統胚の後肢ではこのような状態は観察されなかった。また、Sim1発現細胞をニワトリ胚の肢に移植し、神経配向パターンの変化を調べると、移植した細胞群に向かって軸索が伸長している様子が観察された。その頻度は、コントロール実験としてSim1非発現細胞を移植した場合よりも高いことも確かめられた。これらの解析により、Sim1発現細胞に軸索を誘引する働きがある可能性が示唆された。Sim1の関与をより直接的に証明するためには、今後Sim1の異所的強制発現実験や機能抑制実験をおこない、神経配向パターンに変化が生じるかどうかを検証する必要がある。
以上の研究成果の一部を論文としてまとめ、Nature Communications誌に発表した。
