Outline of Annual Research Achievements

本年度は，DNAによる細胞サイズ小胞型分子ロボットの核となるボディの構築と形成メカニズムの解明及び反応系とのカップリングによる分子ロボットへの動的機能の付与について研究を行った．DNAによる細胞サイズ小胞型分子ロボットのボディの構築は，ハイブリダイゼーションすることによってゲル化する３つ塩基配列が異なるDNAを正電荷脂質に被われた細胞サイズの油中水滴(脂質膜小胞)内に封入，油中水滴の界面においてDNAの動的な自己組織化によって形成される．この形成過程において，ハイブリダイゼーションのためのアニーリングにおける冷却速度を制御することで，中空構造のDNAスフィア，さらには DNAスフィア中にGap構造が見られるDNAスケルトンの2種類のボディを作製することに成功，構造形成を制御することにも成功した．構造形成のメカニズムについても，形成過程のリアルタイム観察と解析，シミュレーションを用いて明らかにした．また，DNAスケルトンに外部から刺激を加えることで，構造を保持したままサイズを収縮させることができることを見出し，ダイナミクスを操作することが可能であることを示した．現在，DNAの塩基配列中にRNAを転写できる配列を付与，反応系とのカップリングによる分子ロボットへの動的機能の付与についても進行中である．
さらには，DNAナノテクノロジーの技術を応用して，DNAオリガミを用いた研究についても進行中である．今後，DNAの塩基配列等を様々に組み替えることで，センシング機能やアクチュエータ機能を有する小胞型DNA分子ロボットの開発に発展させたいと考えている。
