Outline of Annual Research Achievements

前年度までに、HP1はAurora B/CPCの機能を亢進させることで正確な染色体分配に寄与していることを明らかにした。また、がん細胞ではHP1結合量が減少しているためにAurora B/CPCが機能低下し、それによって染色体不安定性が引き起こされていることを突き止め、これらの研究成果をDevelopmental Cell誌に掲載することができた。また今年度は、前年度までに報告した結果に基づき、がん細胞におけるAurora Bの機能低下メカニズムの見解についてCell Cycle誌に掲載することができた。
今年度は、がん細胞においてHP1結合量が低下する要因について検討した。前年度までに、ヒト正常細胞にがん化シグナルを導入して人為的に形質転換させた細胞ではHP1のCPCへの結合量が有意に低下することを見出していた。多くのがん細胞および人為的に形質転換させた細胞では、染色体の数に異常が生じている（染色体異数化）ことが知られている。そこで、ヒト正常細胞に染色体を1本追加導入させた染色体異数化細胞を人為的に樹立しHP1のCPCへの結合量を調べたところ、染色体異数化細胞ではHP1のCPCへの結合量が低下していることを発見した。これらの発見から、染色体数のバランス異常がHP1の結合量を低下させる要因である可能性が示唆された。染色体数の異常は細胞内タンパク質恒常性を変化させることが知られているため、今後はタンパク質恒常性の変化がHP1-CPC結合に影響するかどうか解析を進める必要がある。
