Research Abstract

単純ヘルペスウイルス(HSV)は少なくとも74種の遺伝子をコードするが、その半数以上は培養細胞での増殖には必須ではないアクセサリー遺伝子である。本研究はHSVの増殖、病原性発現におけるアクセサリー遺伝子の役割、作用機構を明らかにしようとするものである。
UL14遺伝子産物は、VP26やUL33を核内に輸送する機能を有すること、分子シャペロン機能をもつことなどを明らかにしてきた。今回、UL14発現細胞(14/HEp-2)の性状について非発現細胞と比較検討したが、14/HEp-2は、様々なアポトーシス誘導刺激に対して抵抗性になっていることが判明した。さらにHSV感染細胞においてもUL14がanti-apoptoticに作用している可能性が示唆された。UL3遺伝子産物はセリン/スレオニンプロテインキナーゼ(PK)でアポトーシス抑制作用をもつことを明らかにしてきた。中枢神経系のHSV感染におけるUS3の役割を知るために脳定位固定装置を用いたマイクロインジェクション法によりマウス嗅球へUS3欠損ウイルス(LIBRI)を接種し、野生株US3レスキュー株と比較検討した。その結果、HSV感染による中枢神経細胞のアポトーシスの抑制にもUS3が関与していることが示唆された。UL51遺伝子産物は、感染後期に合成され、パルチミン酸による修飾を受けるリン酸化粒子構成蛋白質であり、種々のゴルジマーカーと共局在することを明らかにしてきた。2種類のUL51欠損ウイルス(UL51Δ)を作製したが、いずれも野生株と比べてかなり小さなplagueを形成した。電顕による観察では、粒子成熟のレベルで阻害されていることが示唆された。UL56遺伝子産物はC末端アンカー型type II膜蛋白質であること、ゴルジ体や初期エンドゾームに分布することを報告してきた。Yeast two hybrid法による解析から、UL56はKinesin super familyに属するKIFIAのC末端部分と相互作用している可能性が示唆され、これはGST-pull down法によっても確認された。
