Research Abstract

Toll-like receptor (TLR)のアダプターについて以下の点を明らかにした。
1.2種のアダプター、MyD88,TICAM-18TRIF)がシグナル系の選別に直接関与する:MyD88はNF-κBを主に、TICAM-1はIRF-3を主に活性化する。
2.MyD88経路はバクテリア産物に応答しやすいがTICAM-1経路はウィルス産物に応答しやすい。
3.TICAM-1活性化はTLR3,TLR4によって活性発動する。
4.TLR3はTICAM-1を直接リクルートし、活性化するが、TLR4はLPSなど限られたリガンドがTLR4にTICAM-2/TICAM-1複合体アダプターをリクルートしてIRF-3を活性化する。
5.ウィルス感染はTLR-3-TICAM-1以外にTICAM-1下流のkinaseを直接活性化するかTICAM-1経路に入る副経路を使う。ともにIRF-3の活性化に結果する.
6.TICAM-1経路はIRF-3→IFN-βを誘導するので強力な抗ウィルス免疫を起動する。
7.TICAM-1経路はウィルス起因性を含めて各種がんにも抗がん作用を発揮する。
8.TICAM-1の下流はTANK-IKKi/β、TBK-1などと云われているが我々はこれらと異なったデータも得ている。
9.ウィルスの標的分子もこれらが主だが、ウィルス種によってはPKRや他の分子も標的にしうる。
以上の結果(公表文献参照)からTICAM-1経路とウィルス標的分子の同定を種々のウィルス特異的に推進中である。また各TLRについてmAbを作製したので他のTLRについても分子複合体解析(ウィルス感染VS非感染細胞)を行う予定である。
