痛覚回路可塑性のin vivoパッチクランプ解析

Research Abstract

炎症モデル脊髄後角表層、膠様質で観察した、脳由来神経栄養因子(BDNF)による興奮性シナプス伝達の増強作用に、如何なる受容体や細胞内経路が関与するか解析した。炎症初期のモデルから脊髄スライス標本を作製しBDNFを投与すると自発性EPSCの発生頻度が著明に増大した。この作用はチロシンキナーゼinhibitorのK252aによって抑制された。また、TTX存在下ではBDNFによる発生頻度の増大が同様に観察されたが、lidocaine存在下では抑制された。また、PLCのinhibitor存在下でも発生頻度の増大が見られた。これらの結果から、BDNFはPLCを介さずに、神経終末部に発現するTrkB受容体を介してTTX非感受性Naチャネルを活性化し、グルタミン酸の放出を促進する事が示唆された。
次に、BDNFの抗体を用いて炎症初期に観察された興奮性シナプス伝達増強作用を抑制し、脊髄痛覚回路の可塑性発現を阻止できるか否か解析した。炎症後1週間を経過したモデルでは、正常膠様質に入力するAδ線維とC線維に加えてAβ線維由来の単シナプス性EPSCが約30%以上の細胞で観察された。炎症モデルに抗BDNF抗体を投与した動物で解析を行うと、Aβ線維由来の単シナプス性EPSCの入力は減少し、正常と同様のシナプス入力に戻った。行動実験においても抗BDNF抗体を投与した動物では、機械的痛みに対する閾値が有意に増大し、アロディニアの程度が減弱した。以上より、BDNFは脊髄痛覚回路の可塑性発現に関与し、抗BDNF抗体は炎症に伴う痛覚回路へのAβ線維の入力を阻止することが明らかになった。
