Research Abstract

mRNA翻訳抑制作用をもつrapamycinの細胞内標的蛋白質mTOR(mammalian target of rapamycin)は巨大なSer/Thr kinaseである。mTORは、細胞周囲の栄養環境の変化、たとえばアミノ酸濃度の変化を感知して下流分子p70 S6 kinase(p70S6k)などをリン酸化し、蛋白質合成を制御する。Rapamycinは、細胞内でFKBP12と複合体を形成し、この複合体がmTORに結合することによりmTOR機能を阻害するといわれているが、その詳細は不明である。我々は、mTORと複合体を形成する蛋白質raptor(regulatory associated protein of mTOR)を同定した。Raptorは、mTORとその基質であるP70S6kなどの両方に結合し、mTORが基質をリン酸化するための足場を提供するスキャフォールド蛋白質として機能している。本研究では、mTORシグナルの分子基盤の解明を目指し、プロテオミクス的手法によりmTOR/TSC/Rheb関連蛋白質に結合する分子の候補を探索した。その結果、複数の分子が同定されている。興味深いことに、候補分子の1つは、PKB/Aktのリン酸化モチーフを持ち、アミノ酸依存性にこのモチーフ上のリン酸化が変動することが確認された。この分子の同定は、mTORシグナル系の新たな発見につながる可能性を秘めている。この分子以外の候補分子についても精力的に研究を推進しており、本研究の進展が、新たな翻訳調節機構の解明につながるものと期待される。
