動物卵受精による母性mRNA翻訳活性化の分子機構

Research Abstract

アフリカツメガエル(以下、ゼノパス)の受精と初期発生におけるRNA結合タンパク質heteronuclear ribonucleoprotein K(hnRNP K)の機能に焦点を充てた解析を行なった。ゼノパス卵hnRNPK(以下、xhnRNP K)は分子サイズ47kのタンパク質として発現し、大部分は細胞質分画に、一部はポリソーム分画に存在していた。ポリウラシルRNAの固定されたビーズを用いたアフィニティー沈降実験から、xhnRNP KにRNA結合能があることが示唆された。受精後5分から20分の初期胚において、xhnRNP Kは一過的にチロシンリン酸化されていることがわかった。ゼノパス卵で受精直後に活性化されるチロシンキナーゼSrc(xSrc)の阻害剤を注入しておいた卵では、xhnRNP Kの受精依存的なチロシンリン酸化が阻害された。xSrcの活性化を伴う人為的卵活性化処理によってもxhnRNP Kのチロシンリン酸化は誘導された。一方、未受精卵においてxhnRNP Kがセリン/スレオニンリン酸化されていることが分かった。このリン酸化は受精後20分過ぎには低下し、分裂前期の胚において再び一過的な上昇を示した。同じリン酸化の上下動パターンはp42 MAPキナーゼにも見られた。HEK293細胞を用いた強制発現実験と卵抽出液を用いたインビトロリン酸化実験において、xSrcあるいはp42 MAPキナーゼによるxhnRNP Kのリン酸化の再構成に成功した。xhnRNP KのポリウラシルRNA結合能はチロシンリン酸化により低下し、セリン/スレオニンリン酸化により上昇することが示された。以上から、ゼノパス卵xhnRNP KはxSrcおよびp42 MAPキナーゼによるリン酸化の標的としてRNA機能、特に母性mRNA翻訳制御に関与することが示唆された(投稿準備中)。
