Research Abstract

植物細胞の「安定な細胞極性」の成立と維持において基盤となる細胞壁の構築に関わる遺伝子群について、軸性形成の各局面での発現様式を包括的に解析し、各細胞型・組織固有の軸性形成に係わる細胞壁構築遺伝子セットを特定し、それらの発現統御に係わる転写制御システムを分子解剖することを目指し研究を進め以下の成果を得た。
根に特異的な発現パターンを示すAtXTH18のmRNA発現をAtXTH18のプロモーターを用いたRNAi法により特異的に抑制したところ、表皮細胞の伸長成長が抑制され、根の伸長が著しく抑制されることを見いだした。
根で特異的な発現パターンを示すAtXTH12の機能をT-DNAタグライン(xth12-1)を用いて解析し、この遺伝子の欠損変異が根の伸長抑制を引き起こすことを示した。
AtXTH27が葉の維管束形成過程において、発生段階依存的な発現パターンを示すことを見いだした。pAtXTH2::GUSの発現解析から、この遺伝子は葉の維管束、特に前形成層細胞に特徴的な発現パターンを示した。この遺伝子の欠損した変異体(xth27-1,xth27-2)では、ロゼット葉の維管束の形成不全および機能不全をおこし、葉の間抹消維管束周辺の葉肉組織の壊死および三次維管束の篩部機能不全を伴った。
シロイヌナズナとイネのゲノム情報を利用した、両者の細胞壁遺伝子群に関する包括的なinsilico比較ゲノム解析を行った。この結果を基にして、イネOsXTHファミリー29遺伝子の包括的な発現解析を行い、OsXTH19のみがイネ地上部(葉と節間)の伸長組織で特異的且つ顕著な発現を示すことを明らかにした。
