Research Abstract

マウスES細胞はサイトカインLIFによってその全能性が維持される。一方、サルやヒトなど霊長類のES細胞はLIFには反応せず、マウス胎児線維芽細胞(MEF)の抽出物によってその全能性が維持される。我々はMEF由来の分子で霊長類ES細胞の全能性維持を可能にする分子を同定するためにMEF由来のcDNAライブラリーを作製して、カニクイザル由来ES細胞の未熟性を維持する因子の発現クローニング法を試みた。それに加えて候補分子を絞りES細胞維持活性を有する分子を捜す実験も並行して行った。
COS7細胞による発現クローニング法でES細胞の未熟性維持を指標にMEFのライブラリーをスクリーニングしたが単独でES細胞を維持できる因子を同定することはできなかった。一方、検索した候補分子のうちWnt3aにES細胞の維持を支持する活性があることが判明した。しかしながらWnt3a単独では長期間未熟性を維持することは不可能であった。また、マウスES細胞もある程度Wnt3aにより維持されることが判明したが、Wnt3a単独で維持した場合、ALP陽性など未熟性の指標となるマーカーはある程度保たれても、マウスを作る能力は急速に失われた。また、Wnt3aの精製度が上がると逆にES細胞維持能を失う傾向にあった。このことは、Wnt3aによるES細胞維持活性には何らかのco-factorの存在が必要であることを示している。以上の結果はES細胞の未熟性維持にはマウスにおいても霊長類のES細胞においてもMEF由来の複数の分子が必要であることを示している。
