Research Abstract

ヒト臍帯血や骨髄には、様々な組織へと分化し得る能力をもつ細胞が存在することが現象的に知られている。これらの組織に存在する造血幹細胞に関しては、細胞表面抗原の組み合わせにを用いたFACS解析により細胞1個レベルの精度で同定されており、研究の進展には目をみはるものが多い。しかし、各種組織へと分化し得る細胞の同定に関しては、ほとんど解明が進んでおらず、現在問題とされている組織幹細胞の持つ多分化能、可塑性の機構解明への大きな障害となっている。我々はヒト細胞の生着が許容できるNOD/SCID/γ_c^<null>マウスを開発しているが、このマウスとヒト臍帯血細胞移植との系を組み合わせることにより、各種組織へと分化し得る細胞の峻別化が可能となり、組織幹細胞の多様性および可塑性の解明に向けて貢献できることが期待される。本研究において、ヒト臍帯血CD34陽性細胞をNOD/SCID/γ_c^<null>マウスに移植、生着を確認した後、抗Fas抗体やCCI4により肝障害を引き起こすことにより、ヒト細胞由来肝細胞が生成されることを見出した。免疫組織染色によりヒト肝細胞におけるヒトアルブミン産生が確認されるのに加えて、マウス血清中においてもヒトアルブミンの存在が確認され、機能的にもヒト臍帯血由来肝細胞が作動していることが明らかとなった。現在、ヒト臍帯血より肝細胞への分化機構について、検討を加えている。
