Research Abstract

蛋白質は一般に疎水部を水から避ける形で折り畳まれるが、疎水的な内部に水分子が存在する場合もある。これら内部結合水の中には単に空間を充填するだけでなく、機能に重要な役割を演ずる水分子の存在が考えられている。本研究では、濃度勾配に逆らってイオンを輸送する分子ポンプ蛋白質の機能発現過程における内部結合水の役割を明らかにするため、光駆動プロトンポンプであるバクテリオロドプシンなどを対象として、低温赤外分光を用いた研究を行っている。本年度は以下の成果を得た。
バクテリオロドプシンの活性中心を構成する2つのアスパラギン酸(Asp85,212)を非解離性のアミノ酸に置換するとプロトンポンプ活性がなくなる。一方、クロライドを結合するとAsp85の変異体はクロライドポンプに機能転換する一方、Asp212の変異体はプロトンポンプ機能が復活することが知られているが、そのメカニズムは不明であった。我々は昨年、クロライドポンプに転換したD85変異体の構造解析を報告したが、それに続いてD212変異体の赤外分光解析を行った。その結果、シッフ塩基の水素結合アクセプターである水分子の位置にクロライドが結合するD85変異体とは異なり、D212変異体ではシッフ塩基-水-Asp85という水素結合ネットワークを再構成するようにクロライドが活性中心近傍に結合することが明らかになった。強い水素結合を形成した水分子は、ハライドが結合したD212変異体にのみ観測され、ロドプシンのプロトンポンプ機能とも完全に対応している。これ以外にも、バクテリオロドプシンのAsp115の変異体や海洋性バクテリアのプロトンポンプ蛋白質であるプロテオロドプシン(PR)に対して赤外分光解析を行い、特異的な水素結合構造が機能発現に重要な役割を担っていることを明らかにした。
以上の成果として、2007年に11編の原著論文を世に出すことができた。また、平成19年度には15件の招待講演を含む46件の学会発表を行った。
