Research Abstract

蛋白質と周囲の水分子との相互作用、特に反応途中における相互作用のダイナミクスを時間分解で明らかにするための研究を行った。また、蛋白質の反応を物理化学的に研究するため、新しい検出手法の開発を行い、本年度は以下のような成果を得た。
1.我々の開発した時間分解拡散係数法を、青色受容タンパク質であるYcgFに適用し、BLUFドメインに関与する反応を時間分解で明らかにした。その結果、光励起に伴い、数十マイクロ秒での構造変化とダイマー化反応の存在を示した。吸収法では、このダイマー化という劇的な反応ダイナミクスでも全く検出されないことから、本手法は反応機構を明らかにする非常に有力なものであることがわかった。
2.植物の持つ青色光センサーであるフォトトロピンについて、Phot1LOV2ドメイン単体(LOV2試料)とそれにlinkerを付随させたもの(LOV2-linker試料)を用いて、その反応を時間分解TG測定により研究した。得られた拡散信号の形や強度に観測時間依存性が見出され、光照射により誘起された拡散係数変化を伴う蛋白質全体の構造変化が観測されていると結論した。LOV2試料では、光照射に伴いダイマー化と解離反応が起こっていることが示された。また、LOV2-linker試料では、LOV2試料に比べてより大きな拡散係数変化が光励起後1msの時定数で観測された。これは光照射により1inker部分のヘリックスが壊れ、溶媒との相互作用が強まり、拡散係数が大きく変化したためであり、1inker部分のヘリックス崩壊という大きな構造変化が光照射後1msで誘起されていると結論づけられた。
3.PYPと呼ばれる光センサータンパク質の構造変化について、拡散係数の観点からそのダイナミクスを調べた。その結果、N末端のヘリックスが壊れる速度は、従来考えられていたよりも1桁速いことが明らかとなった。
