Research Abstract

多細胞生物の組織構築,形態形成は高度な細胞間の相互作用によって規定される.細胞間相互作用の素過程としてのシグナル伝達は初期発生から個体レベルでの恒常性の維持まで,広範な生命活動を実現する基本反応のひとつであり,組織・個体レベルの機能と,細胞レベルの機能の接点としてきわめて重要な位置を占める.オルガネラダイナミクスによる発生プログラムの制御を明らかにするため,オルガネラ形成分子の欠損マウスを解析した,酵母の液胞形成タンパク質Vam2は液胞膜上での膜融合装置として機能する,マウスのVam2ホモログ,mVam2を同定し,遺伝子破壊マウスを作成した.mVam2欠損胚では,visceral endoderm(VE)のエンドソームリソソームが断片化し,酵母のclass II vam破壊株と同様な形態を示す. m Vam2欠損マウス胚はBMPシグナルがectopicに活性化されており,原腸陥入は開始するものの,予定神経領域外胚葉で細胞死が誘導され,頭部形成が強く阻害されて胎性7.5日以降の発生が進まない.野生型胚ではBMP受容体がVEに特徴的なリソソーム,apical v acuoleに運ばれるが,m Vam2変異株はapical vacuoleを欠いており,受容体の分解コンパートメントへの輸送が起きていない.これらの観察結果は,BMP受容体のリソソームへの膜輸送による不活性化が,マウス初期発生におけるBMPシグナルの空間的な分布の形成,さらには初期発生プログラムの実現に極めて重要な素過程であることを示している.
