Research Abstract

X染色体上にGFP遺伝子が挿入されたマウスを用いると着床前の初期胚の段階で簡単に性の分別を行うことができる。この系を利用して作製・収集したXX型の精原細胞やXXY型精巣において過剰に発現している遺伝子群の発現状況を詳細に検討したところいずれも胎生18.5日目から急激に発現が始まっていることを認めた。またプロゲステロン投与により分娩を遅らせても同時期からの発現上昇と、XX精原細胞の消失時期に変化がないことが確かめちれた。これらは分娩という環境の変化にかかわらず遺伝子発現のカスケードによりXX型細胞の消失につながっていることを示すものであった。またXX型セルトリ細胞の能力について検討を加えたところ、XX型セルトリ細胞は移植した精原細胞を精子にまで分化させる力を持っていることが見出せたが、継続した精子形成支持能はないことが観察された。この原因を探らたところ、自己免疫疾患に似た抗体が産生されていることを発見した。精原細胞が消失してしまう原国としてセルトリ細胞が担っている免疫的なバリアの形成がXXタイプのセルトリ細胞では十分に機能せず、そのために精原細胞が非自己として免疫的に攻撃を受けている可能性が示された。
一方、着床前の胚の遺伝子発現を雌雄で比べることによりRhox5遺伝子がインプリント遺伝子であることを示していたたが、今回は同様にX染色体上にあるFth117クラスター遺伝子がインプリントされている可能性が高いことを見つけた。しかしながらクラスターであるために相互に分別した発現解析が難しく、その役割を明らかにするためにノックアウトでこの領域全体を除去すべくターゲティングベクターを作製した。
