Research Abstract

脳が機械的損傷やウイルス感染などを受けると活性化アストロサイトが出現するが、その活性化機構や機能的な役割はまだよく分かっていない。 昨年度までの報告で、我々が作成した、SNAP-25変異マウスの脳で、生後10週齢頃から抗GFAP抗体で強染色される活性化アストロサイトが、大脳皮質や海馬を中心に出現することを見いだした。本年度はSNAP-25変異マウスの脳で見られる活性化アストロサイトの性質を明らかにするため、様々な抗体を用いて免疫組繊化学解析を行った。イムノプロット解析から変異マウスの脳ではBDNF発現も上昇することが明らかとなったため、抗BDNF抗体を用いて免疫組繊化学解析を行ったところ、活性化アストロサイトに強い陽性反応を見いだした。さらに様々な抗体を用いて網羅的染色を行った結果、GDNFやSynaptogyrinに対しても陽性反応を示すことが明らかとなった。興味深いことにBDNF陽性の活性化アストロサイトは海馬や大脳皮質に存在しているのに対し、Synaptogyrin陽性細胞は海馬に、GDNF陽性細胞は海馬歯状回に限定して見いだされた。これらの結果からアストロサイトには、活性化の度合いや機能の違いによる多様性が存在することが考えられた。さらに活性化アストロサイトからの神経栄養因子の分泌機構を明らかにするため、ラット由来のグリオーマ細胞にBDNFを発現させたところ、細胞内顆粒に貯蔵され、Ca2+依存的な機構で放出されることが明らかとなった。
