Research Abstract

髄鞘形成グリアと軸索との相互作用を明らかにする目的で,下記の実験を行った.
髄鞘主要糖脂質スルファチドの欠損マウスでは,末梢神経ランビエ絞輪に巨大なミトコンドリア様小胞が集積し,軸索膜が膨隆する.各種マーカーを用いて解析した結果,COXIVやMnSOD陽性のミトコンドリアの絞輪への集積が明らかになった.このような変化は髄鞘形成直後の生後7日目から存在し,成長と共に増加した.これらはATP合成酵素COXVやチトクロームc陽性で機能を有すると推測された.ミトコンドリアの巨大化や集積が輸送の異常によるか軸索内環境の変化によるか解析中である.
Annexin IIは,末梢神経髄鞘のparanodeおよびSchmidt-Lanterman切痕に集積する.末梢神経の脱髄モデルマウスでは脱髄部分から周囲にかけてAnnexin IIが増加し、phospho1ipase A2(PLA2)と共局在することから,PLA2抑制を介して脱髄を防ぐ可能性が示唆された.
脱髄や軸索-髄鞘間の相互作用に関与する分子の解析には,2次元電気泳動(2-DE)と質量分析を組み合わせたプロテオミクス解析が有用である.しかし,髄鞘タンパク質は高塩基性や高疎水性分子が多く,通常の2-DEでは解析が困難なため,昨年度までに陽イオン界面活性剤16-BACを用いた2-DEの有用性を報告した.本年度は新たに陽イオン界面活性剤CTABが疎水性タンパク質の可溶性に非常に有効であることがわかった.これらの2-DE各々の特長を生かして組み合わせることで,より網羅的なタンパク質の解析が可能になる.実際に16-BACを用いて髄鞘画分に濃縮される高分子としてMyosin 1Dを同定した.これは髄鞘形成時期に一致して増加し,白質線維に局在することが明らかになった.現在,髄鞘形成や維持に対するMyosin 1Dの作用を解析中である.
