Research Abstract

本年度(平成18年4月〜平成18年7月)の研究実績
1 走査型蛍光X線顕微鏡システムの構築
これまで開発を進めてきた、ナノ集光システムの後段に、既存のXYステージと蛍光X線ディテクターを装着させ、走査型の蛍光X線顕微鏡システムを構築した。XYステージには、0.01μm分解能のステージを用いた。蛍光X線ディテクターからの波形分布を、各ポイントで取得可能なシステムとし、複数の元素マップを取得することを可能とした。SPring-8のBL29XULにおいて、テストパターンによるシステムの動作確認と、細胞内の元素分布の観察を行った。テストパターンはFIB (Focused Ion Beam)により作製した。顕微鏡システムの空間分解能のテストを行った結果、30nmの分解能でテストバターンに書かれていた文字を画像化することができた。
細胞観察では、細胞内の核やミトコンドリアの観察において、同時に複数の元素分布を可視化することに成功した。
2 ブロジェクション顕微鏡システムの予備検討
ブロジェクション型顕微鏡システムとしては、将来的な発展性を考慮すると、近年注目をあびているX線回折顕微鏡の導入が不可欠であり、検討を開始した。本手法は、X線の透過強度分布から、集光点近傍のサンプル内の電子分布を求めるものであり、原理的にナノメートルの空間分解能を持つ顕微鏡手法として有力である。今年度は、位相回復法と呼ばれる数学的手法に基づくブログラムを開発した。そして、ミラー集光光学系においても、透過強度分布からサンブルの電子構造を求めることが可能であることがわかった。
