Research Abstract

1)出芽酵母染色体DNA複製開始前複合体(pre-RC)の形成反応(複製開始部位への逐次的なORC→Cdc6p→Cdt1p→Mcm2-7p複合体のLoading反応)をAFMを用い詳細に研究した。又、高度に精製した蛋白複合体を使って形成させたpre-RCにS-期に同調した出芽酵母細胞から調整した核粗抽出液を加え、試験管内において複製開始部位から特異的に開始するDNA複製反応系を確立した。
2)出芽酵母染色体DNA複製に必須なGINS, RPA, Polεポロ酵素複合体とssDNAを鋳型として使って、Leading鎖合成反応を再構成し,詳細に解析した。
3)アフリカツメガエル卵粗抽出液の染色体DNA複製反応に必要なGINS, Cut5,Polεホロ酵素複合体,Cdc45p等の蛋白複合体を精製し、GINS-Polεポロ酵素複合体,Cdc45p-Polεホロ酵素複合体,GINS-cdc45p-Cut5-Polεホロ酵素複合体形成反応を試験管内で再構成した。更に、Polεポロ酵素複合体を除去したアフリカツメガエル卵粗抽出液とDNAポリメラーゼ活性ドメインを持たないPolεホロ酵素複合体を用い、完全なPolεホロ酵素複合体が染色体複製反応に必須であることを証明した。
4)前年度に同定したマウス5番染色体上に存在する複数の複製開始部位のより詳細な解析をPCR法とDNAマイクロアレイを使用して行った。
5)出芽酵母染色体DNA複製開始に必須なCdc7-Dbf4複合体によるチェックポイント蛋白Rad53pのリン酸化による活性化機構を詳細に解析した。又、精製したRad53pが持つ一本鎖DNA部分に特異的に結合する反応を詳細に解析した。
