Research Abstract

クルマエビの眼柄ペプチドホルモンのうち血糖上昇ホルモン族ペプチド(血糖上昇ホルモン(CHH)および脱皮抑制ホルモン(MIH))について以下の結果を得た。
1.CHHのすべての分子が、MIHが有しているN末端から12番目のGlyを有していない。このGlyがCHH活性にとって不要なものか、あるいは逆に活性を低下させるのかを明らかにするために、CHHのうちのひとつCHH-IについてN末端から12番目のGlyを挿入した組み換え体を作製し、そのCHH活性を調べた。その結果、挿入したものは挿入しないものの約1/10の活性しか示さなかった。このことから、このGlyが入らないことが高活性を維持するのに重要であることがわかった。
2.CHHの立体構造はこれまでMIHの立体構造に基づいてモデリングで推定されているだけであった。CHHの実際の立体構造を解析するために^<13>Cおよび^<15>NでダブルラベルしたCHH-Iの組み換え体を大腸菌で発現した。この分子は封入体に回収されたので、いったん還元したあとNi樹脂上でリフォールディング反応を行ったが、ラベルしないときは進んだ反応がラベルした分子では同じようには進行しないことがわかった。現在、この反応の収率を上げることを検討中である。
3.MIHとMIH受容体の結合について、標的器官であるY器官の膜画分を用いてスキャッチャード解析を行った結果、解離定数Kd=4.76x10^<-10>M、最大受容体結合量Bmax=5.51x10^<-12>Mであると推定された。これらの値は、他の甲殻類で報告されている値とほぼ同程度であった。
