Research Abstract |
1) Recombinant human bone morphogenetic protein-2 (rhBMP-2) with polylactic acid-polyglycolic acid copolymer/gelatin sponge (PGS) carrier was applied on two-wall- and three-wall surgically created periodontal defects on adult dogs. The histometric results suggested that rhBMP-2 enhanced bone formation in three-wall-periodontal defects. 2) Horizontal circumferential periodontal defects were surgically created around the root of premolar bilaterally in six beagle dogs. An elastic ligature was placed tightly around the root at the base of the defect in order to inhibit coronal migration of cells from intact periodontal ligament tissue. Enamel matrix derivative (EMD) was applied in the experimental side and carrier alone was applied in the contra-lateral side as control. rhBMP-2/PGS was applied in the experimental and control sites. The distance between the bottom of the defect and apical terminal of ankylosis was significantly greater in EMD group compared to control. 3) Adenovirus with core binding factor alpha-1 (Cbfa1) gene adsorbed to porous beta-tri-calcium phosphate block was applied on horizontal circumferential periodontal defects in adult dogs. As all blocks were exposed to oral cavity in healing period, it was impossible to evaluate the effect of Cbfa1 on periodontal regeneration in this study. 4) Experimental periodontitis was induced on the mesial side of maxillary canines in six adult dogs. Calcium phosphate cement (CPC) was injected in the defects. Non-grafted defects on the contralateral side served as controls. Twelve weeks after surgery, periodontal tissue healing was evaluated histometrically. New bone, new cementum, and new connective tissue attachment and adhesion were significantly enhanced in the experimental sites.
