Research Abstract |
It is well known that some physiological functions are elicited via non-Selective cation channels. The function and the distribution of vanilloid receptor TRPV1(transient receptor potential vanillbid-1) in rodent were studied in order to clarify whether TRPV1 functions as non-selective cation channels. As a result of this project, I got the following results. (1)Peripheral neuron cell : Capsaicin, an agonist of TRPV1, induced transmitter release from PC12 cells, a rat pheochromocytoma cell line, through TRPV1. (2)Gastric function : Capsaicin protects gastric mucosa through activation of TRPV1 in rat gastric corpus lesion and antral ulcer models. In addition, it induced gastric acid secretion through central TRPV1 in anesthetized rats. (3)Distribution of TRPV1 in stomach : Numerous TRPV1-immunoreactive axons were also found around arterioles in the submucosal layer. A large number of nerve fibers were seen in the smooth muscle layer and in the myenteric nerve plexus. (4)Exploration of natural occurring product acting on Ca^<2+> channel blocker-resistant Ca^<2+> influx : Mesaconitine, a major constituent of aconite, acts on non-selective cation channel, leading to release of nitric oxide from the endothelium in isolated rat aorta. Gingerol, a major constituent of ginger, acts on non-selective cation channel, leading to enhancement of peristaltic movement of intestinal tract.
