Research Abstract

Research Background : Mesenchymal stem cells in bone marrow have been known to differentiate into bone and cartilage cells in vitro culture system. Human cord blood is abundant with the mesenchymal stem cells in comparison with adult bone marrow. We analyzed the possibility of differentiation and proliferation of mesenchymal stem cell(MSC) in vivo, which derived from cord blood stem cell transplantation donor.
1)Analysis of bone and cartilage cells derived from donor cells (cord blood stem cell)
An infant with familial hemophagocytic lymphohistiocytosis(FHL) received a cord blood stem cell transplantation. After the engraftment of the donor cells occurred, osteolysis was found in his skull and bone biopsy was done. We analyzed whether the specimen include the donor (cord blood stem cell) derived MSC or not, using the PCR method targeting the G08062 on the chromosome 20. The results revealed the mixed chimerism with 70% of donor derived cells. Then we prepared the single cell PCR method to analyze the origin of single cell extracted by the microdissection method, but it was difficult to get the enough DNA from the bone tissue for the analyses.
2)Analysis of bone marrow cells derived from donor cells (cord blood stem cell)
A 6-year-old boy with refractory Burkitt lymphoma received a cord blood stem cell transplantation from HLA 2 loci mismatched donor. We analyzed how the chimerism varied in the different cell lineage of bone marrow after the transplantation, using the PCR method targeting the microsatellite markers on the chromosome 18,20,22 and HLA disparity. Unfortunately, the bone marrow relaplse was found soon after the transplantation, so the results revealed the 100% of recipient derived cells.
