In this study, we demonstrated that multiple cord blood transplantation may be useful alternative transplantation for the patients who do not find suitable donor. 1)Lethally irradiated BALB/c (H-2d) mice were transplanted bone marrow cells derived from B6 (H-2b) and C3H (H-2k) mice. Both of donor cells were successfully engrafted to show mixed chimerism. Similarly, while B6 and C3H newborn liver cells as a model of cord blood, both cells were engrafted. 2)One or two months after transplantation, lymphocytes were reconstituted, and CD4+CD25+ cells were detected in flowcytometer. It was remained whether there are relationship between the number of CD4+CD25+ cells and the rate of engraftment for B6 and C3H cells or not. 3)Four months after transplantation, spleen cells were used to make mixed lymphocyte reaction(MLR) to get uptake of radioactive 3H-thymidine. MLRs for both types were low uptake, suggesting that multiple donors' tolerance were induced. 4)To further investigation, we have remained a subject that what is the role of regulatory T cells, CD4+CD25+ cells, for engraftement or long term chimerism in multiple cord blood transplantation.
