Research Abstract

Sialoglycoconjugates is thought to play an important role in tumor metastasis and progression. This study involved histochemical analyses of human colorectal cancer and metastatic liver cancer tissues to detect tumor-associated sialoglycoconjugates and to clarify the relationship between their histochemical expression and the metastatic potential of the tumor.
Human colorectal cancer and metastatic tissues were subjected histochemical staining using sialic acid-binding lectins and anti-KL-6 mucin monoclonal antibody. Relationship between the staining characteristics and clinicopathological parameters were statistically analyzed. Aberrant expression of sialoglycoconjugates was detected in colorectal cancer tissues by means of lectin-histochemical staining. The high level staining was related to aggressive tumor behavior such as lymphatic metastasis. On the other hand, KL-6 mucin was detected in colorectal cancer tissues but not in normal tissues surrounding the tumors. The expression was significantly frequent not only in cases of lymph node metastasis but also in those of liver metastasis. In addition, KL-6 antigen was detected in metastatic liver cancer tissues with high frequency (53/56, 95%).
In conclusion, the present results indicate that tumor-associated sialoglycoconjugates appear in human colorectal cancer and metastatic liver tissues and suggest that the aberrant expression of sialoglycoconjugates, especially KL-6 mucin, is closely related to unfavorable behavior such as liver metastasis of colorectal cancer.
